Liposomes are very popular drug carriers for oral administration due to their excellent biocompatibility and versatility. However, there are some issues concerning liposome functionality and stability, such as an increase in the leakage of the entrapped contents, and lipid oxidation. The lipid bilayer is a dynamic barrier that forms a closed internal structure; therefore, a disturbance of the lipid bilayer structure may cause these problems. There is only one report on the monitoring of a liposome bilayer using Raman scattering, where no significant changes in the Raman intensity were observed. However, coherent Raman scattering (CRS) has a high spatial resolution and is thus suitable for detecting liposome defects. In this study, a liposome consisting of dimyristoyl-phosphatidylcholine (DMPC) and palmitic acid (PA) was used to study the effect of Cu(II) and H2O2 on the integrity of the lipid bilayer. An increase in the PA content of the lipid bilayer was observed after Cu(II) and H2O2 exposure. Moreover, the lipid oxidation levels of the liposomes were determined using CRS. The CRS intensity of PA in the liposomes gradually decreased in response to Cu(II) and H2O2 exposure. As a result, CRS has the potential to be used to monitor the stability of the lipid bilayer in liposomes.Not every sales rep will be able to do these full time.
